Required Materials
- Lyophilized peptide vial
- Sterile diluent appropriate to your assay (for example sterile water or bacteriostatic water, per your protocol)
- Sterile graduated syringe or calibrated pipette
- 70% isopropanol wipes
- Sterile gloves
- A disinfected work surface (a biosafety cabinet or laminar flow hood is preferred)
Step-by-Step Procedure
- Prepare sterile materials. Gather all items and disinfect the work surface with 70% isopropanol.
- Calculate the diluent volume. Volume (mL) = peptide quantity (mg) divided by target concentration (mg/mL). For example, 5 mg of peptide in 2 mL of diluent gives 2.5 mg/mL.
- Disinfect the vial stoppers. Wipe both stoppers with an alcohol wipe and allow 30 seconds to evaporate.
- Draw the diluent. Draw the calculated volume and remove air bubbles.
- Add diluent along the vial wall. Tilt the vial and add the diluent slowly down the inner wall. Do not direct the stream onto the lyophilized cake, which can damage the peptide.
- Dissolve by gentle swirling. Swirl gently; do not shake. Dissolution usually takes from 30 seconds to a few minutes. The solution should be clear with no visible particles.
- Label and store. Label with the compound name, final concentration, date and your initials. Store according to the product documentation and your protocol.
Storage Considerations
Reconstituted peptides are generally less stable than the lyophilized form. Stability depends on the specific peptide, diluent and conditions; refer to the product documentation and your own stability data. Aliquoting can limit repeated freeze-thaw cycles.
Critical Points
- Do not heat the peptide or use sonication to speed dissolution unless your validated protocol allows it.
- Do not add diluent directly onto the lyophilized powder.
- Avoid repeated freeze-thaw cycles.
- Inspect for particles after dissolution; persistent turbidity may indicate a problem. Contact us with your batch number.
